  设计和合成了一系列与阿司咪唑有关的化合物，目的是确定它们的抗疟原虫活性。阿斯咪唑结构的几种修饰，即除去4-氟苄基和/或4-甲氧基苯乙基部分，取代苯并咪唑支架的苯环，取代4-氟苄基中的氟原子，以及改变4探索了氨基哌啶部分。使用ItG菌株体外评估这些化合物的抗疟原虫活性表明，阿斯咪唑及其某些结构相似的衍生物具有IC 50 值在纳摩尔范围内，并且对中国卵巢仓鼠（CHO）细胞表现出对寄生虫的毒性，选择性高达200。苯并咪唑部分中第2位存在仲环胺并在4和5位被氯取代有两种修饰可以产生基于阿斯咪唑的有效和选择性抗疟药。

  Astemizole is a H-1-antagonist endowed with antimalarial activity, but has hERG liabilities. Systematic structural modifications of astemizole led to the discovery of analogues that display very potent activity as inhibitors of the growth of the Plasmodium parasite, but show a decreased hERG inhibition, when compared to astemizole.
