The enantioselective hydrolysis of 17 racemic α-substituted arylacetonitriles by Rhodococcus sp. CGMCC 0497 is described. The corresponding (R)-amides and (S)-acids were obtained with excellent enantiomeric excess in most cases. Known (R)-1-aryl-alkylamines of the formula ##STR1## in which R.sup.1 and Ar have the meanings indicated in the description, are prepared by a new process, which comprises a) reacting a racemate of an ethyl or methyl 2-aryl-alkanoate of the formula ##STR2## with ammonia in the presence of a lipase which is suitable for the cleavage of esters, in the presence of a diluent, b) separating off from the reaction mixture the resulting (R)-2-aryl-alkanamide of the formula ##STR3## and then reacting it with sodium hypochlorite or sodium hypobromite in the presence of water.

Nach einem neuen Verfahren lassen sich (R)-1-Aryl-alkylamine der Formel
in welcher
R1 und Ardie in der Beschreibung angegebenen Bedeutungen haben,
herstellen, indem man
a) Racemate von 2-Aryl-alkansäure-ethylestern bzw. -methylestern der Formel
mit Ammoniak in Gegenwart von Lipasen, die zur Spaltung von Estern geeignet sind, in Gegenwart eines Verdünnungsmittels umsetzt,
b) aus dem Reaktionsgemisch die entstandenen (R)-2-Aryl-alkansäure-amide der Formel
abtrennt und dann mit Natriumhypochlorit oder Natriumhypobromit in Gegenwart von Wasser umsetzt.

Microbial hydrolysis as a potent method for the preparation of optically active nitriles, amides and carboxylic acids
Many kinds of aromatic nitriles have been hydrolyzed to afford the corresponding amides and carboxylic acids, by the aid of enzyme system of Rhodococcus butanica. This enzymatic hydrolysis can be successfully applied to the kinetic resolution of α-arylpropionitriles, resulting in the formation of (R)-amides and (S)-carboxylic acids.
