1-substituted 2-benzylaminobenzimidazole derivatives: compounds with H1-antihistaminic activity

The preparation of 1-substituted 2-benzylaminobenzimidazole derivatives 13-24 is described. Although these compounds have a different structure from the general structure I of antihistamines, they showed some antihistaminic activity (pA2:5-7) when tested in vitro on guinea pig ileum. The introduction of an additional basic centre as in the Mannich bases 25-27 did not cause any improvement of activity. All the compounds tested proved to be inactive in in vivo induced passive cutaneous anaphylaxis (PCA) and cutaneous vasopermeability in rats.

Benzimidazole compounds that increase the rate of SOS-mediated nucleotide exchange on Ras by binding to a functionally relevant, chemically tractable pocket on the SOS protein, as part of the Ras:SOS:Ras complex.
The direct N-alkylation of 2-aminoimidazoles to give the corresponding 2-(N-alkylamino)imidazoles was accomplished using alcohols as alkylating agents in the presence of a [Cp*IrCl2]2/K2CO3system. The iridium-catalyzed regioselective reaction is simple, efficient, general, and environmentally benign.

A novel photoarylation of aminoacids and peptides is described, which tolerates the presence of water. Irradiation of Boc‐protected aminoacids in the presence of N‐protected 2‐azidobenzimidazoles leads to selective arylation of carboxy termini or side chains. The new reaction also works for peptides.
