The best of both worlds: Screening of a substrateactivityscreening (SAS) library first for inhibitors and then for substrates generates complementary data sets. Combined, these provide coherent SAR data that can be used both for fragment‐based construction of noncovalent small molecules and for the production of inhibitors with covalent warheads.
