    The present disclosure provides compositions and methods for the delivery of immune cells to treat un-resectable or non-resected tumor cells and tumor relapse. The compositions comprise (i) a structure comprising an injectable polymer or scaffold comprising pores; (ii) lymphocytes disposed within the structure, (iii) at least one lymphocyte-adhesion moiety associated with the structure; and (iv) at least one lymphocyte-activating moiety associated with the structure, and optionally an immune stimulant.

    The present invention relates to scaffolds that can physically guide cells, e.g. neurons, while best matching the material properties of native tissue. The present invention also relates to methods of generating such scaffolds, and for the use of such scaffolds, e.g. in spinal cord and peripheral nerve injury repair. The methods of the present invention include a uniquely controlled freeze casting process to generate highly porous, linearly oriented scaffolds. The scaffolds of the present invention not only comprise a highly aligned porosity, but also contain secondary guidance structures in the form of ridges running parallel to the pores to create a series of microstructured and highly aligned channels. This hierarchy of structural guidance aligns and guides neurite outgrowth down the channels created by the ridges, and keep neurites from branching perpendicular to the inter-ridge grooves.

    The present invention provides methods for producing a product of one or more enzymatic pathways. The pathways used in the methods of the invention involve one or more conversion steps such as, for example, an enzymatic conversion of guluronic acid into D-glucarate (Step 7); an enzymatic conversion of 5-ketogluconate (5-KGA) into L-Iduronic acid (Step 15); an enzymatic conversion of L-Iduronic acid into Idaric acid Step 7b); and an enzymatic conversion of 5-ketocluconate into 4,6-dihydroxy 2,5-diketo hexanoate (2,5-DDH) (Step 16). In some embodiments the methods of the invention produce 2,5-furandicarboxylic acid (FDCA) as a product. The methods include both enzymatic and chemical conversions as steps. Various pathways are also provided for converting glucose into 5-dehdyro-4-deoxy-glucarate (DDG), and for converting glucose into 2,5-furandicarboxylic acid (FDCA). The methods also involve the use of engineered enzymes that perform reactions with high specificity and efficiency. Additional products that can be produce include metabolic products such as, but not limited to, guluronic acid, L-iduronic acid, idaric acid, glucaric acid. Any of the products can be produced using glucose as a substrate or using any intermediate in any of the methods or pathways of the invention.
