    Connecting Proteins by Design. Cross-Linked Bis-Hemoglobin
    摘要:
    A new type of multifunctional reagent creates a specific connection within and between two hemoglobin tetramers, resulting in a cross-linked bis-tetramer ("CLBT"). The tetrafunctional reagent (N,N'-5,5'-bis[bis(3,5-dibromosalicyl)isophthalyl]terephthalamide, DBIT) was prepared by conversion of the corresponding tetraacid to the tetrakis(3,5-dibromosalicylate). Deoxy hemoglobin reacts with DBIT to give a cross linked bis-tetramer as the major product. Patterns in tryptic digests reveal that there are modifications of amino groups at beta -Lys-82 and beta-Val-1, indicative of a structure in which each tetramer is cross-linked between these positions. The cross-linked bis-tetramer has a decreased affinity for oxygen and very low cooperativity in oxygen binding. The properties of the material provide insights into the nature of protein-protein interactions.

    Synthesis of Analogues of Congo Red and Evaluation of Their Anti-Prion Activity

    No cure as of yet exists for any of the transmissible spongiform encephalopathies. In this paper, we describe the synthesis of analogues of Congo red and evaluation against a cellular model of infection, the SMB (scrapie mouse brain) persistently infected cell line, for their ability to inhibit the infectivity of the abnormal form of prion protein (PrP-res). The compounds have also been tested for their ability to inhibit the polymerization of PrPC by PrP-res. A number of analogues showed inhibition of PrP-res infectivity at nanomolar concentrations. Several analogues show promise; the most active compound, 2a, inhibits the formation of PrP-res in SMB cells with an EC50 of 25-50 nM.

    Two multifunctional Mn(II) metal–organic frameworks: Synthesis, structures and applications as photocatalysis and luminescent sensor

    Two Mn(II) metal-organic frameworks, [Mn2L(1,10-phen)(2)]center dot DMF center dot 0.5H(2)O (1) and [Mn(1,10-phen)(SO4)(H2O)(2)] (2) (H4L = bis-(3,5-dicarboxyphenyl) terephthalamide) have been synthesized from Mn(II)/H4L system under different pH values conditions. The structures of the two MOFs were determined by single crystal X-ray diffraction analysis and characterized by elemental analysis, IR spectroscopy and thermogravimetric analysis. In compound 1, organic ligand (L4-) adopt six-dentate modes to link Mn(II) ions into 2D layer structure. For 2, only sulfate ion and 1,10-phen molecule link Mn(II) ions into 1D chain structure under higher pH value. The results demonstrate that pH values play an important role in the construction of the two MOFs. We also studied the photophysical properties and applications of the two MOFs as photocatalysts and fluorescence sensor materials. The photocatalysis results indicate that the two MOFs are stable and good candidates for degradation of RhB under UV light irradiation. And also, 1 has the higher photocatalytic activity than that of 2. In addition, the fluorescence sensing experiment demonstrates that 1 have significant selectivity in detection of aromatic amines and metal ions. All these results demonstrate that MOF-based materials can be used as multifunctional materials for photocatalytic degradation of organic dyes and detection of aromatic amines/or metal ions.
