  一系列的手性合成 六氢异苯并呋喃(HIBF) 核苷是通过立体定义的（顺式异构体）糖基序16与适当的甲硅烷基化碱基进行糖基化来实现的。所有核苷类似物均以 52-71% 的产率获得，作为 α- 和 β-异头产物的混合物，增加了可用于筛选的新型核苷的范围。新型双环 HIBF 核苷的结构是通过 β-HIBF 胸腺嘧啶类似物22b 的单晶 X 射线结构确定的。此外，这些核苷的糖构象被确定为 N 型。在合成的新型 HIBF 核苷中，测试了 25 种化合物作为人外周血单核 (PBM) 细胞中 HIV-1 的抑制剂，发现 7 种化合物具有活性 (EC 50= 12.3–36.2 μM）。其中六种化合物是具有 β-HIBF 的嘌呤类似物肌苷在所有测试的化合物中，类似物22o是最有效的 (EC 50 = 12.3 μM)。之间惊人的相似去羟肌苷 (滴滴) 和22o可以解释有效的抗 HIV 活性。
