The present invention provide water soluble polyfluorenes functionalized with glucuronic acid useful in sensing bilirubin in aqueous medium and process for preparation thereof. The invention further deals with detecting bilirubin in human serum samples in the range from normal (<25 μηioI/L˜1.2 mg/dL) human bilirubin level to jaundiced bilirubin level (>50 μηioI/L˜2.5 mg/dL). This is a fluorescence turn-off mode of detection where blue fluorescence of polymer quenches and becomes colorless. The water soluble polyfluorenes functionalized with glucuronic acid can detect free bilirubin in the range from 1×1O″4M to 1×10′7M moles in aqueous and buffer media as a change in the fluorescence signal.

A bilirubin-specific enzyme composition comprises a fungal enzyme having a protein content which, in the presence of a bilirubin-containing aqueous liquid having a pH of about 7.4 and a temperature of about 37°C, degrades at least about 0.02 micromole of bilirubin per minute per milligram of protein. The enzyme may be extracted from mushrooms, and may have the ability to generate hydrogen peroxide.
The enzyme composition may be employed in wet and dry analytical methods and dry analytical elements either in assays for bilirubin or to suppress bilirubin interference in assays for other materials.

A highly sensitive fluorimetric method for analysis for bilirubin contained in an aqueous liquid comprises:
(a) contacting together the aqueous liquid and an interactive. polymeric mordant for bilirubin to mordant bilirubin;
(b) thereafter, or during step (a), subjecting the mordanted bilirubin to activating radiation effective to excite the mordanted bilirubin and produce detectable fluorescence;and
(c) detecting this fluorescence to obtain a determination relating to the presence and/or concentration of bilirubin in the test sample.
The method may be conducted in solution or in a dry test element.
