The in vitrodistribution and fate of [14C]diethylmalonate and [14C]diisopropylfluorophosphate were evaluated on normal and heat-treated pigskin. The extent of hydrolysis from the skin surface, skin, and receptor fluid was determined. A significant skin-mediated hydrolysis (15-35% of applied dose) was observed for diethylmalonate in normal skin, but not in heat-treated skin. These results indicated
New phosphinylalkanoyl prolines which have the general formula ##STR1## wherein R.sub.1 is lower alkyl, phenyl or phenyl-lower alkyl; R.sub.2 is hydrogen, phenyl-lower alkyl or a metal ion; R.sub.3 is hydrogen or lower alkyl; R.sub.4 is hydrogen, lower alkyl, phenyl-lower alkyl or a metal ion; and N is 0 or 1, Are useful as hypotensive agents.
New phosphonoacyl prolines and related compounds have the general formula ##STR1## wherein R.sub.1 and R.sub.2 each is hydrogen, lower alkyl, lower alkenyl, unsustituted or substituted phenyl-lower alkyl, or a metal ion; R.sub.3 is hydrogen or lower alkyl; R.sub.4 is hydrogen, lower alkyl, phenyl-lower alkyl or a metal ion; and N is 0 or 1. These compounds are useful as hypotensive agents.
