    Novel and practical preparation of α-fluoro-functionalized esters from fluoroiodoacetates
    摘要:
    The addition reaction of fluoroiodoacetates 2 to various electron-rich alkenes 3 initiated by iron powder in dry THF at 70-80 degrees C gave 1 : 1 adducts 4 in good yields. A variety of functionalities in the alkenes such as trimethylsilyl, alkoxy, acetoxy, hydroxy and ester could be tolerated under the reaction conditions. Reduction of the adducts 4 with Zn-AcOH in ethanol or Zn-NiCl2 . 6H(2)O in moist THF was readily accomplished, and the overall procedure was amenable to a convenient one-flask procedure, Treatment of fluoroiodoacetates 2 with electron-deficient alkenes 7 in the presence of an Fe-CrCl3 . 6H(2)O-bpy bimetal redox system in ethanol at 70-80 degrees C resulted in the formation of iodine-free 1 : 1 adducts 8 in moderate to good yields. It is proposed that the addition reactions of fluoroiodoacetates 2 to electron-rich and electron-deficient alkenes proceeded through a single-electron-transfer mechanism.

    The title compound 2 can be alkylated with a wide range of alkyl halides and Michael acceptors. Subsequent thermal elimination of phenyl sulfinic acid 3 leads to α-fluoro-α,β-unsaturated ethyl carboxylates 5 and 10, an important class of intermediates for fluorine containing biologically active compounds.

    Anodic methoxylation of various types of N-(fluoroethyl)amines, ArRNCH(2)R(f) (R(f) = CF3, CHF2, CH2F, etc.) has been systematically studied and it was found that a methoxy group was exclusively or preferentially introduced into the position a to the fluoromethyl (R(f)) group, depending on the R(f) and R groups. The effect of the R(f) group on the promotion of the anodic alpha-methoxylation decreased in the order CF3, CHF2, and CH2F. This remarkable promotion effect and unique regioselectivity can be explained mainly in terms of the alpha-CH kinetic acidities of the cation radicals formed by one-electron oxidation of the amines. The alpha-methoxylated products are highly useful precursors for the construction of carbon-carbon bonds alpha to the trifluoromethyl and difluoromethyl groups, which is difficult by other methods.
