The first enantiospecific total synthesis of (3S,5S)-hydroxypiperazic acid 1 is described. The synthesis begins fromD-mannitol and exploits a tandem electrophilic hydrazination/nucleophilic cyclisation reaction to assemble the hexahydropyridazine ring system.
