Synthesis and structure-activity relationship of substituted 1,2,3,4-tetrahydroisoquinolines as N-type calcium channel blockers
摘要:
Voltage Activated Calcium Channel (VACC) blockers have been demonstrated to have utility in the treatment of pain and stroke. A series of aminomethyl substituted isoquinolinol derivatives with potent functional activity for N-type VACC's have been identified. Their synthesis and preliminary pharmacology are discussed herein. (C) 1998 Elsevier Science Ltd. All rights reserved.
aqueous acetonitrile solutions, containing compounds that consist of active methylene moieties tethered to aryl-substituted alkenes along with sodium hydroxide and 9-cyanophenanthrene. When the substrate contains ethyl cyanoacetate as an asymmetric active methylene moiety, the reaction proceeds in a highly diastereoselective manner.
combination of “TiCl2(cat)” and Zn (cat=catecholate) facilitates the homolytic cleavage of “non-activated” alcohol C−O bonds. All aliphatic primary, secondary and tertiary alcohols serve as good substrates. This method was applied to radical conjugate addition reactions successfully, and mechanistic studies indicate that alkyl chlorides are not intermediates. The active species is a 1 : 2 complex of alkoxide
