Deoxyribonucleoside‐modified squaraines were synthesized by Sonogashira coupling reactions using an unsymmetrical, terminal alkynylated benzothiazolium squaraine dye. These non‐natural nucleosides exhibited fluorescent ‘turn‐on’ properties in viscous conditions with an enhancement of >300‐fold.
