    Die Erfindung betrifft ein Präparat zur Applikation von Wirkstoffen in Form kleinster, insbesondere mit einer membranartigen Hülle aus einer oder wenigen Lagen amphiphiler Moleküle bzw. mit einer amphiphilen Trägersubstanz versehenen Flüssigkeitströpfchen, insbesondere zum Transport des Wirkstoffes in und durch natürliche Barrieren und Konstriktionen wie Häute und dergleichen. Das Präparat weist einen Gehalt einer randaktiven Substanz auf, der bis zu 99 Mol.-% des Gehaltes dieser Substanz entspricht, durch den der Solubilisierungspunkt der Tröpfchen erreicht wird. Das Präparat eignet sich zur nichtinvasiven Verabreichung von Antidiabetica, insbesondere von Insulin. Die Erfindung betrifft außerdem ein Verfahren zur Herstellung solcher Präparate.

    A sorbitol kinase having at least the following physicochemical properties: 1) enzymatic action: utilizing mainly ATP, but not substantially phosphoenolpyruvate, as the phosphate donor catalyzing the following reaction: sorbitol + ATP → sorbitol 6-phosphate + ADP;(2) substrate specificity: having the substrate specificity for sorbitol; (3) N-terminal amino acid sequence: containing the following sequence: Met-Arg-Ile-Gly-Ile-Asp-Leu-Gly-Gly-Thr-Lys-Thr-Glu-Val-Ile-Ala-Leu-Ser-Glu-Gln; (4) thermostability: stable at least up to 60 °C when a 100 mM tris-HCl buffer (pH: 8.5, containing 0.5 U/ml of sorbitol kinase) is heated for 15 minutes at various temperatures and the residual activity is measured.

    The present invention relates to a probe compound that can comprise any substrate or metabolite of an enzymatic reaction in addition to an indicator component, such as, for example, a fluorescence dye, or the like. Moreover, the present invention relates to means for detecting enzymes in form of an array, which comprises any number of probe compounds of the invention which each comprise a different metabolite of interconnected metabolites representing the central pathways in all forms of life. Moreover, the present invention relates to a method for detecting enzymes involving the application of cell extracts or the like to the array of the invention which leads to reproducible enzymatic reactions with the substrates. These specific enzymatic reactions trigger the indicator (e.g. a fluorescence signal) and bind the enzymes to the respective cognate substrates. Moreover, the invention relates to means for isolating enzymes in form of nanoparticles coated with the probe compound of the invention. The immobilisation of the cognate substrates or metabolites on the surface of nanoparticles by means of the probe compounds allows capturing and isolating the respective enzyme, e.g. for subsequent sequencing.
