The syntheses of optically pure 4,4-dimethyl-L-glutamic acid as well as (2S,4R)- and (2S,4S)-4-hydroxy-4-methylglutamic acids have been achieved by transamination of the corresponding 2-oxo-4,4-dimethyl- and rac-2-oxo-4-hydroxy-4-methylglutaric acids using glutamic oxalacetic transaminase (GOT).

Synthesis of stereoisomers of 4-bromo- and 4-iodo-N-phthaloyl-glutamates based on the bromination of N-phthaloylglutamic acid followed by separation of diastereoisomers is described.
Efficient bienzymatic cascades combining aldolases and α-transaminases were designed for the synthesis of γ-hydroxy-α-amino acids. These recycling cascades provide high stereoselectivity, atom economy, and an equilibrium shift of the transamination. L-syn or anti-4-hydroxyglutamic acid and D-anti-4,5-dihydroxynorvaline were thus isolated in 83-95% yield in one step from simple substrates.

Four optical isomers of γ-hydroxyglutamic acid were prepared in pure states by opticalresolution of two racemic diastereoisomers through brucine salt of N-benzyloxycarbonyl amino acid or strychnine salt of N-benzoyl derivative. Configurations of α- and γ-asymmetric carbon atoms of these isomers were estimated from contribution to molecular rotation and character of taste exhibition. These determinations
