    Guest:host interactions of lidocaine and prilocaine with natural cyclodextrins: Spectral and molecular modeling studies

    Inclusion complex formation of two local anesthetics drugs (lidocaine (LC) and prilocaine (PC)) with α- and β-cyclodextrins (CDs) in aqueous solution were studied by absorption, fluorescence, time-resolved fluorescence and molecular modeling methods. The formation of inclusion complexes was confirmed by 1H NMR, FTIR, differential scanning calorimetry, SEM, TEM and X-ray diffractometry. Both drugs formed 1:1 inclusion complex and exhibit biexponential decay in water whereas triexponential decay in the CD solution. Nanosized self-aggregated particles of drug: CD complexes were found by TEM. Both experimental and theoretical studies revealed that the phenyl ring with the amide group of the drug is encapsulated in the hydrophobic CD nanocavity. Investigations of energetic and thermodynamic properties confirmed the stability of the inclusion complexes. van der Waals interactions are mainly responsible for enthalpy driven complex formation of LC and PC with CDs.

    The present invention relates to the use of cyclic oligosaccharides as chemical complexants of lipofuscin bisretinoids (A2E) to prevent and treat eye (i.e., retinal or macular) disease. Monomeric, dimeric, multimeric, or polymeric oligosaccharide rings act as pharmacologic agents to prevent and treat ophthalmologic disorders triggered by the accumulation of lipofuscin in the retinal pigment epithelium (RPE), which occurs as a consequence of either genetic disorders, such as Stargardt Disease (SD) and Best Disease (BD), or aging, such as Age-Related Macular Degeneration (AMD), or other diseases, such as retinitis pigmentosa, and cone-rod dystrophy.

    Aspects of the present disclosure include a system for analytical sample preparation. In some embodiments, the system includes a container having disposed therein an analytical sample treatment composition including α-cyclodextrin and/or an α-cyclodextrin co-polymer. Also provided is a method of reducing matrix effects in an analytical sample. In some embodiments, the method includes: contacting a sample including a matrix-interfering agent and an analyte with a cyclodextrin composition to produce a matrix-cyclodextrin complex, wherein the cyclodextrin composition comprises an α-cyclodextrin and/or an α-cyclodextrin co-polymer; separating the complex from the contacted sample to produce a matrix-reduced composition; and detecting the analyte in the matrix-reduced composition. Kits and compositions for use in the subject systems and methods are also provided.

  • Molecular Recognition by Self-Assembled Monolayers of Cyclodextrin on Ag

    Self-assembled monolayer (SAM) of a thiolated cyclodextrin (CD) derivative was prepared, and stereoselective inclusional complexation of optically active azo dyes (p-or o-methyl red conjugates with chiral I-phenylethylamine, p-or o-MR-PEA) with both free CD and the SAM of the CD derivative on silver surface was investigated by using resonance Raman (RR) and surface-enhanced resonance Raman spectroscopy (SERRS), respectively. Characteristic molecular vibrations of functional groups of MR-PEA were used to describe the complexation. To our knowledge, this is the first report which compares the complexations of free and surface-confined CD by Raman spectroscopy. Association constants for the complexation (K) were determined from the relative intensity of Raman scattering of the dye complexed with CD. The K values for the complexation between MR-PEA and free CD determined by Raman spectroscopy were quite similar to those measured by W-visible spectroscopy, which confirmed the validity of Raman spectroscopy for the analysis of the inclusion phenomena. Though the K values of the SAM system were much larger than that of the free system, similar stereoselectivity was observed in both of the systems. The R enantiomer of o-MR-PEA was mon preferentially included both by free and surface-confined CD than the S-enantiomer, whereas a slight selectivity was observed for p-MR-PEA. The importance of the position of optically active carbon atom with respect to the azobenzene moiety, which penetrated into the cavity of CD, was suggested. Our findings show that the enantioselectivity of CD is preserved after immobilization onto solid surface.
