The present invention relates to novel hydroxyethylamine compounds having Asp2 (β-secretase, BACE1 or Memapsin) inhibitory activity, processes for their preparation, to compositions containing them and to their use in the treatment of diseases characterised by elevated β- amyloid levels or β-amyloid deposits, particularly Alzheimer's disease.

2-Formyl-malondialdehyde () reacts at 22°C in a type of Diels-Alder reaction with reversed electron demand with the enol-ethers () – () and () – () within a frew hours to give the dihydropyrans () – () and () – (). Thio-enol-ethers may also be employed in this cycloaddition.

In this paper we present a simple and efficient two-step procedure for the preparation of triformylmethane from bromoacetic acid and dimethylformamide, synthesis of some triformylmethane derivatives of preparative importance, evidence for an intramolecular hydrogen bond in triformylmethane, based on its 1H-NMR spectra measured in deuterochloroform in the presence of trichloroacetyl isocyanate, and comments on some recent erroneous statements1 concerning the IR spectra of triformylmethane.
An Improved Isolation of Triformylmethane (TFM): Properties and Preparation of Some Derivatives

We present a straightforward method for the isolation of the title compound, triformylmethane (TFM), describe some of its physical properties, and present some evidence for its versatility as a synthetic building block for heterocyclic compounds. The structure of TFM appears to be medium-dependent as determined by infrared and nuclear magnetic resonance spectroscopy.

Six new compounds were obtained by condensation of triformylmethane with some aminoadamantane derivatives. The study also includes 7-amino-1,3,5-triazaadamantane.
