Two unsaturated peptide substrates were each cyclised by the enzyme isopenicillin N synthase (IPNS) from Cephalosporium acremonium CO 728 simultaneously to both desaturated and hydroxylated β-lactam products whose hydroxy groups have been shown to derive their oxygen atom from the cosubstrate dioxygen; a working hypothesis involving iron–oxo species is proposed to explain this phenomenon.

A series of unsaturated tripeptides have been synthesised and evaluated as substrates for isopenicillinNsynthetase; variations in the C-terminal amino acid have led to a structure reactivity profile which indicates a stereochemical dependence on the balance between desaturative vs. hydroxylative ring closures.

extract of Cephalosporiumacremonium, no conversion to active antibiotics was observed; however on co-incubation with the Arnstein tripeptide (ACV) (2), strong inhibition of the conversion of ACV to isopenicillin N was observed.
