Boronic ester-based self-healing hydrogels formed by using intermolecular B-N coordination

The copolymer poly(acrylamide-co-2-acrylamido glucose) (PA) and poly(acrylamide-co-N-acryloyl-3-aminophenylboronic acid-co-N-(3-dimethylaminopropyl) acrylamide) (PB) were separately prepared by radical polymerization, and the hydrogel was readily constructed by simple mixing the two kinds of copolymers. The amine-containing monomer, N-(3-dimethylaminopropyl) acrylamide was incorporated into the polymer chain to enhance the stability of the complex between boronic acid and glucose units under physiological condition. The mechanical properties of the hydrogel can be readily engineered by adjusting the pH, solid content, and [phenylboronic acid]/[glucose] ratios. Moreover, the hydrogel possesses excellent intrinsic self-healing ability. Benefitting from the reversible property of the boronic ester, the hydrogel can be dissociated by adding competitive sugar molecules, which makes it easy to remove from medical gauze or skin. Additionally, cell cytotoxicity studies revealed that the hydrogel has good biocompatibility. With its superior properties, this dynamic phenylboronic ester-based hydrogel has great potential to be a component of new generation of wound dressings.
A water-soluble protein conjugate modified by saccharides is covalently bound to a carbohydrate backbone via a saccharide group. Such protein conjugates modified by saccharides can be produced in a simple way by polymerizing a vinylsaccharide and conjugating in a known way the poly(vinylsaccharide) obtained in this way with the protein, which is preferably an enzyme. The protein conjugates according to the present invention are stable over long time periods at a high enzymatic activity and also in aqueous solution and are thus particularly suitable for use in test kits.
The liquid, polymer-containing coating materials of this invention comprise a polymer of a polymerizable, nitrogen-containing, hydrophilic monomer, which may give thermoresponsive properties, and which is copolymerized with a hydrophobic, polymerizable siloxy-containing monomer, in a solvent system of a volatile hydrophobic (non-polar) liquid that is non-stinging to a user. The material forms a coating or bandage in the form of a film when applied to a surface or the skin of a user.

The object of the present invention is to provide a conjugate of a therapeutic agent for joint diseases and hyaluronic acid, a hyaluronic acid derivative or a salt thereof which can retain the therapeutic agent for joint diseases in joint cavities. According to the present invention, there are provided a conjugate of at least one therapeutic agent for joint diseases and hyaluronic acid, a hyaluronic acid derivative or a salt thereof; a method for preparing the above described conjugate which conjugate comprises binding a site of the therapeutic agent for joint diseases (for example, a matrix protease inhibitor) which does not affect the activity of a therapeutic agent to a carboxyl group, a hydroxyl group or a functional group at the reducing end of hyaluronic acid, a hyaluronic acid derivative or a salt thereof by direct chemical reaction or via a spacer; and a pharmaceutical composition containing the above described conjugate.
