    A labeled nucleic acid probe comprising (a) a nucleic acid component, (b) a nucleic acid-binding ligand photochemically linked to the nucleic acid component, and (c) a label chemically linkedto the nucleic acid-binding ligand. The label can be a specifically bindable ligand such as a hapten or biotin, an enzyme such as a ß-galactosidase or horse radish peroxidase, a fluorescent radical, a phycobiliprotein, a luminescent radical, or a radioisotope. The probe can be used in assays of nucleic acids, taking advantage of the ability of the nucleic acid component to hybridize.

    A nucleic acid hybridization assay wherein the hybrid formed between the polynucleotide sequence to be detected and the probe is immobilized by binding of an immobilized or immobilizable form of an antibody reagent selective for binding such hybrid. Preferred antibody reagents are selective for DNA·RNA or RNA·RNA hybrids or for intercalated duplexes. The immobilized hybrid is detected preferably by use of a labeled form of a second such antibody reagent or a labeled form of the probe. The method dispenses with the need to immobilize either the probe or sample nucleic acids and permits hybridization to proceed between the sequence to be detected and the probe in solution, resulting in rapid rates of hybridization.

    Nucleic acid hybridization assay methods and reagent systems for detecting a particular polynucleotide sequence in a test medium. An aggregate is formed in the assay reaction mixture comprising intercalation complexes between a nucleic acid intercalator and double stranded nucleic acid associated with the hybridization product of the sequence to be detected and a nucleic acid probe sequence. Hybridization of the probe with the sequence to be detected can then be determined by addition of an antibody, or a fragment thereof, capable of binding with the intercalation complexes in the formed aggregate and measuring the antibody or fragment thereof which becomes bound to such intercalation complexes associated with hybridized probe. In one preferred embodiment, this method eliminates the need to chemically modify the probe in order to form a labeled reagent. In another embodiment, the method provides an advantageous method for labeling the probe by chemical modification.

    @ A nucleic acid hybridization assay wherein the hybrid formed with the probe has binding sites for two specific binding reagents, one of which comprises a first label and the other a second label, interaction of the first and second labels providing a detectable response which is measurably different when the two labeled reagents are both bound to the same hybrid compared to when the two labeled reagents are not so bound. The formation of the hybrid assay product brings the two labels within a proximate interaction distance of one another, e.g., as in the cases of sequential catalyst (enzyme) interaction and energy transfer. Since the labels provide a response which is distinguishable when they are associated with a hybridized probe, no separation step is required.

    A nucleic acid hybridization assay involving a labeled probe and formation of a hybrid having epitopes for an antibody reagent. The label provides a detectable response which is measurably different when the labeled probe is comprised in a hybrid that is bound by the antibody reagent compared to when not comprised in such a hybrid. Particularly useful antibody reagents are antibodies such as anti-DNA'RNA, anti-RNA'RNA and antibodies to intercalated duplexes which do not bind substantially to single stranded nucleic acids. Modulation of the label response can be accomplished in a variety of ways such as by steric inactivation or hindrance or by labeling the antibody reagent with a second label which interacts with the first label. The assay is particularly advantageous because no separation of hybridized and unhybridized probe is required.
