  • Molecular classification of psoriasis disease-associated genes through pharmacogenomic expression profiling

    Psoriasis is recognized as the most common T cell-mediated inflammatory disease in humans. Genetic linkage to as many as six distinct disease loci has been established but the molecular etiology and genetics remain unknown. To begin to identify psoriasis disease-related genes and construct in vivo pathways of the inflammatory process, a genome-wide expression screen of multiple psoriasis patients was undertaken. A comprehensive list of 159 genes that define psoriasis in molecular terms was generated; numerous genes in this set mapped to six different disease-associated loci. To further interpret the functional role of this gene set in the disease process, a longitudinal pharmacogenomic study was initiated to understand how expression levels of these transcripts are altered following patient treatment with therapeutic agents that antagonize calcineurin or NF-κB pathways. Transcript levels for a subset of these 159 genes changed significantly in those patients who responded to therapy and many of the changes preceded clinical improvement. The disease-related gene map provides new insights into the pathogenesis of psoriasis, wound healing and cellular-immune reactions occurring in human skin as well as other T cell-mediated autoimmune diseases. In addition, it provides a set of candidate genes that may serve as novel therapeutic intervention points as well as surrogate and predictive markers of treatment outcome.

    A thermosetting epoxy resin composition of one pack type is provided which contains as indispensable constituents (I) an epoxy resin containing more than one adjacent epoxy groups on the average in the molecule and (II) curing amount or curing-accelerating amount of a compound obtained by reacting (a) a specified amine/epoxy adduct with (b) phenolic , resin(s) and/or polyhydric phenol compound(s) in a weight ratio of (a) to (b) of 1/0.1 to 1/0.8. The composition of the present invention has excellent storage stability and it is curable at a relatively low temperature to form a cured product having excellent mechanical strength, adhesiveness and water resistance.

    Disclosed are novel high flexural modulus polyurethane plastics wherein a polyisocyanate and a particular combination of extenders including cycloalkane diol are reacted optionally in the presence of a minor proportion of high molecular weight polyol. The polymers have outstanding resistance to high temperatures thereby expanding their utilities into higher temperature end-use environments than related prior art materials. Certain members of these new polymers are also possessed of outstanding optical clarity which thereby provides a combination of properties heretofore not available in a polyurethane. These polymers are easily melt-fabricated notwithstanding their inherent high temperature properties.

    A process for making the thermoplastic thiourethane-urethane copolymers for use in making optical products comprising forming e.g. a thiourethane prepolymer, preferably by reaction of an aliphatic diisocyanate with a dithiol, and then reacting the prepolymer with a polyurethane-forming monomer which is preferably the combination of a diisocyanate such as methylene bis(phenyl isocyanate) and a polyol such as a diaromatic containing diol such as ethoxylated Bisphenol A or an aliphatic isocyanate reactive monomer such as tricyclodecane dimethanol or 1,4-dimethanol cyclohexane, to form the copolymer product. The polymers have a combination of high refractive index and high Abbe No. The polymers also have high impact strength, high hardness and Tg's above 100°C. It is preferred to make the polymer in an extruder.
