The present invention provides an effective and simple method for selectively acylating insulin or insulin analogues or precursors thereof at a free T-amino group in a lysine amino acid residue in the insulin molecule. The acylating reaction is conducted at mild reaction conditions at neutral pH and at a low water content in the acylation reaction mixture.
