Discovery of imidazo[1,2-b]pyridazine derivatives as IKKβ inhibitors. Part 1: Hit-to-lead study and structure–activity relationship

Imidazo[1,2-b] pyridazine derivatives from high-throughput screening were developed as IKK beta inhibitors. By the optimization of the 3- and 6-position of imidazo[1,2-b] pyridazine scaffold, cell-free IKK beta inhibitory activity and TNF alpha inhibitory activity in THP-1 cell increased. Also, these compounds showed high kinase selectivity. The structure-activity relationship was revealed and the interaction model of imidazo[ 1,2-b] pyridazine compounds with IKKb was constructed. Direct intermolecular C-H arylation of 6-chloroimidazo[1,2-b]-pyridazine in its 3-position was achieved, and the tolerance to reaction conditions in the presence of chloro groups was investigated. Various 3-(hetero)arylimidazo[1,2-b]pyridazines were synthesized in good to excellent yields. This methodology was successfully applied to the synthesis of 3,6-di-and 2,3,6-trisubstituted imidazo[1,2-b]pyridazines
