Ligands with a 3,3-diphenylpentane skeleton for nuclear vitamin D and androgen receptors: Dual activities and metabolic activation

Ligands possessing dual vitamin D-3 (VD3)-agonistic and androgen-antagonistic activities with various activity spectra were prepared based on a substituted 3,3-diphenylpentane (DPP) skeleton. Among the compounds, (R,S)-DPP-1023 [(R,S)-7b] and (S,S)-DPP-0123 [(S,S)-7c] showed the most potent vitamin D-3-agonistic activity [with potency comparable to that of 1 alpha,25-dihydroxyvitamin D-3 (1,25-VD3)] and nuclear androgen receptor (AR)-binding activity (with higher affinity than that of hydroxyflutamide), respectively. Metabolic activation (reduction of the carbonyl group) of pivaloyl analogs [DPP- 1113 (3a), DPP- 10 13 (3b), DPP-0113 (3c), and DPP-0013 (3d)] in HL-60 cells was found to be necessary for binding to nuclear vitamin D-3 receptor (VDR). wherein A represents a hydrophobic unit comprising at least one kind of hydrophobic groups selected from a hydrocarbon group and a heterocyclic group, J represents a connecting group, X-Pro represents a hydrophilic group protected by a protective group Pro which is removable by light exposure, m represents 0 or 1, and n represents an integer of not less than 1.
The protective group Pro may be removable by light exposure in association with the photosensitizer (especially, a photo acid generator), or may be a hydrophobic protective group. The hydrophilic group may be a hydroxyl group or a carboxyl group. The photoactive compound has high sensitivity to a light source of short wavelength beams, for resist application, therefore, the photoactive compound is advantageously used for forming a pattern with high resolution.

The invention relates to a method of preparing silicone copolycarbonates which incorporate both blocky and random substructures. Such silicone-containing copolycarbonates are useful in the preparation of a variety of molded articles such as optical disks. The structure of the compositions prepared by the method of the invention is determined by the method of preparation used and may be used to control physical properties. The copolymers are prepared by forming in a first step an intermediate silicone copolycarbonate possessing either a blocky or random structure and reactive endgroups, and then appending additional structural in a second step which is complimentary to the first step and produces a product silicone copolycarbonate having both random and blocky substructures. The repeat units I and II appended in the second step may be the same or different as those repeat units I and II incorporated in the first step.

A polycarbonate polymer includes: a monomer unit represented by a formula (1A) below; and a monomer unit represented by a formula (2). A haze of a solution that is obtained by dissolving the polycarbonate polymer in THF (tetrahydrofuran) at a concentration of 10 mass % is 5% or less (according to JIS K7105, optical path length of 10 mm).
