    Development of Potent Non-Carbohydrate Imidazole-Based Small Molecule Selectin Inhibitors with Antiinflammatory Activity
    摘要:
    A novel series of non-carbohydrate imidazole-based selectin inhibitors has been discovered via high-throughput screening using a P-selectin ELISA-based assay system. The initial lead 1 had an IC(50) of 17 muM in the P-selectin ELISA; this potency was significantly improved via an extensive SAR exploration. One of the current lead compounds (29) has an IC(50) of 300 nM in a P-selectin ELISA; it also has good activity in P- and E-selectin cell adhesion assays and shows efficacy in vivo. These compounds represent a novel series of sLe(X) mimetics with antiinflammatory activity. Their unique profile supports our interest in their further evaluation as drug candidates for the treatment of inflammation. Herein we describe the syntheses, optimization, and SAR of this series of novel potent selectin antagonists.

    Copolymers comprising: (a) 60 to 99% by weight of at least one monomer selected from the group consisting of N-vinyllactams, N-vinylamides, and mixtures thereof; (1) 1 to 40% by weight of at least one monomer selected from the group consisting of vinyl esters of aliphatic branched C 8 -C 30 -carboxylic acids; (c) 0 to 30% by weight of vinyl acetate; and (d) 0 to 39% by weight of at least one additional free-radically copolymnerizable monomer; wherein the % by weight content of components (a), (b), (c) and (d) totals 100%, and with the proviso that the total amount of component (b) and component (c) combined is 1 to 40% by weight based on the copolymer, are described along with methods of using such copolymers to solubilize substances which are insoluble in water, substances which are only sparingly soluble in water, and combinations thereof.

    The present invention relates to a process for the preparation of N-substituted acrylamides of the general formula CH2═CH—CONHR, wherein, R is alkyl group having carbon 1 to 22 or acyl group having carbon 1 to 18. The process for the preparation of the N-substituted acrylamides comprises reacting acrylamide with alkyl acyl chloride in the presence of Lewis acid catalyst in an organic solvent at a temperature ranging between room temperature to 50° C. for a period ranging between 1 hour to 24 hours.

    This invention describes novel hydrophobically associating ter or tetra polymers which can contain sulfonate functionality which are useful as aqueous fluid rheology or flow modifiers. These high molecular weight water soluble polymers contain both water soluble and water insoluble monomers. The water soluble monomers are N, N dimethylacrylamide acrylamide (DMA) and optionally a salt of an ethylenically unsaturated sulfonic acid (s) or N-vinylpyrrolidinone or a metal acrylate and the water insoluble monomer is a higher alkyl acrylamide (R). The process for their preparation relies on solubilizing the water insoluble monomer into an aqueous micellar solution containing one or more surfactants and the water soluble monomers. The surfactants are specifically selected to enable homogeneous dispersion of the hydrophobic monomer(s) in the presence of anionic sulfonate containing monomers. Redox or free radical initiators are used to copolymerize both the water soluble and water insoluble monomers, forming terpolymers of dimethylacrylamide, alkylacrylamides and acrylamide, and optionally an anionic sulfonate or carboxylate monomer. Aqueous solutions of these hydrophobically associating polymers exhibit enhanced viscosification, reduced salt sensitivity and other desirable rheological properties found useful in a variety of applications.
