There are disclosed two new antibiotics formerly denominated MC916-A and MC916-B and now called neothramycin A and neothramycin B which are potent inhibitors of the growth of leukemia cells, e.g. Leukema L-1210 cells in mice. They are produced by controlled fermentation of Streptomyces FERM-P 2452 (A.T.C.C. 31123).

A one step synthesis of 1,4-benzodiazepines from o-haloanilines and amino acids was achieved by use of palladium catalyzed carbonylation, by which application a synthesis of the model compounds(23a and 23b) of Neothramycin (A and B) was described. An efficient chemoselective reduction of the amide was provided.
An efficient totalsynthesis of neothramycins A (1a) and B (1b) was achieved from the key intermediate (11) which was prepared from o-halogenoaniline derivative (3) and hydroxy-1-proline (4) by use of palladium catalysed carbonylation.
