Nitroxide spin labels can be protected by hydrogenation and alkylation with light‐sensitive groups. Such derivatives are stable under a broad range of critical conditions used in the synthesis of peptides and oligonucleotides. When attached to a nucleoside, standard phosphoramidite chemistry allowed labeling of a short model DNA and recovery of the nitroxide in good yield.
