    Monitoring the DNA by ruthenium complexes of heterocyclic N,S-donor ligands and evaluation of biological activities

    Neutral N,S-donor bidentate ligands have been synthesized and characterized by NMR and IR spectroscopic techniques. The ligands have been used to synthesized ruthenium(II) complexes ([Ru(L-1-L-6)PPh3)(2)Cl-2]) and ruthenium(III) complexes ([Ru(L-1-L-6)PPh3)Cl-3]). Synthesized complexes have been characterized using elemental analysis, UV-Vis spectroscopy, magnetic measurements, LC-MS, and IR spectroscopy. Broth dilution technique was used to investigate antibacterial activity against two Gram-positive and three Gram-negative microorganisms and results show that all complexes are more potent than the respective ligands. UV-Vis absorption titration and viscosity measurements have been carried out to investigate the binding mode and binding strength of complexes with herring sperm DNA. The quantitative binding strength (K (b)) of the complexes was found in the range of 0.521 x 10(5)-2.94 x 10(5) M-1 and indicate intercalative mode of binding. Gel electrophoresis study was carried out to study the cleavage of pUC19 supercoiled DNA. DNA nuclease activity data for the complexes are found higher than respective ligands and metal salt. Brine shrimp bioassay was carried out to perform cytotoxicity study. IC50 values of the complexes were found in the range of 5.69 +/- 1.06-14.62 +/- 0.87 mu g cm(-3).

    Cytotoxic, antibacterial and nucleic acid interaction studies of square planar palladium(II) complexes

    A series of substituted N,O-donor bidentate ligands(L-n) and their square planar mononuclear palladium(II) complexes of the type [PdCl2(L-n)] were synthesized and characterized by elemental analysis, conductivity measurement, TGA, FT-IR, H-1 NMR, C-13 NMR, LC-MS and electronic spectroscopy. Antibacterial activities of the complexes were evaluated against five pathogenic bacteria. The antimicrobial test results reveal that all the compounds have good antibacterial activity against both Gram((+ve)) and Gram((-ve)) bacterial species compared to ligands and metal salt. Interaction of a palladium complex with DNA was investigated by absorption titration and viscosity measurement. Hypochromism and bathochromism were observed as a result of interaction between palladium(II) complexes with DNA. It suggests an intercalative binding mode. Furthermore, viscosity measurements data support the intercalation mode of binding. The palladium(II) complexes were found to promote cleavage of plasmid pUC19 DNA from the supercoiled form to the open circular form.     Novel 3-(4,6-diarylpyrimidin-2-yl)-2-phenylthiazolidin-4-ones are synthesized by the multicomponent cyclocondensation reaction of the appropriate 2-amino-4,6-diarylpyrimidines, benzaldehyde, and thioglycolic acid catalyzed by dicyclohexylcarbodiimide. The synthesized compounds have been characterized by melting point, elemental analysis, MS, FT-IR, H-1 and (13) NMR spectroscopic data.

    Bis(1,3-diketonato)palladium(II) complexes were synthesized by three routes. In route one, sodium hexachlorodipalladate in methanol was treated with the appropriately substituted diaryl-1,3-diketones using sodium carbonate, while in route two substituted diaryl-1,3-diketones were treated with palladium chloride in acetone to afford the desired bis(1,3-diketonato)palladium(II). In route three 1,3-bis(substituted aryl)prop-2-enones were treated with palladium chloride and sodium hydrogen carbonate in THF to provide the oxygen insertion product, bis(1,3-diketonato)palladium(II) complexes. All synthesized compounds were characterized on the basis of spectral and analytical data.
