    The title compounds were synthesized by the efficient route previously explored for the synthesis of enantiomeric pairs of thiolactomycin and its 3-demethyl derivative. These studies were carried out to prove the flexibility of the previously explored synthetic route to natural thiolactomycin (TLM) 1 and to examine the structure–activity relationship on the 5-position of 1. While all of the synthesized congeners lacked in vitro antibacterial activity, these studies led us to find 5-(alk-2-enyl)-TLM (ent-4d) which exhibits mammalian type I fatty acid synthase (FAS) inhibitory activity equal to that of C75, a potent inhibitor reported previously. It was also found that 5-[(E)-cycloalk-2-enylidenemethyl]-TLM (ent-5c) exhibited slightly less potent mammalian type I FAS inhibitory activity than C75.

    cost-effective cis-di­hydroxylation reaction of acrylate derivatives was achieved.     A series of 2-phenyl- and 2-(2-heteroarenyl)-bearing amines possessing 1,2-disubstituted and 1,1,2-trisubststuted alkenes have been evaluated in intramolecular hydroaminations catalyzed by Y[N(TMS)(2)](3) (1(Y)). Aminoalkenes possessing a terminal 2-(5-trimethylsilyl)thienyl group exhibited substantially enhanced reactivity compared to their 2-(phenyl)-containing counterparts. Cyclization efficiencies imparted by the 2-[(5-trimethylsilyl) furanyl] substituent were comparable or only slightly better than those obtained with the simple the 2-(phenyl) group.

    The reaction of chloro, chloromethyl and chlorophenyl carbenoids with ketene alkylsilylacetals has been studied. Excellent yields of cyclopropanation were observed and the unstable chlorocyclopropanone acetals formed were thermally rearranged in high yield into α-substituted α,β-ethylenic esters. This new method for the synthesis of unsaturated esters appeared complementary of the known-ones.
