    A convenient approach for the synthesis of 3-difluoroalkyl phthalides has been developed from phthalaldehydic acids and difluoroenoxysilanes by using relatively inexpensive p-toluenesulfonic acid monohydrate (PTSA) as a catalyst. A series of 3-difluoroalkyl phthalides and cyclic difluoroalkyl ethers were obtained in up to 99% yield. The products obtained could be readily converted into difluoroalkyl

  • Endo-Selective Pd-Catalyzed Silyl Methyl Heck Reaction

    A palladium (Pd)-catalyzed endo-selective Heck reaction of iodomethylsilyl ethers of phenols and aliphatic alkenols has been developed. Mechanistic studies reveal that this silyl methyl Heck reaction operates via a hybrid Pd-radical process and that the silicon atom is crucial for the observed endo selectivity. The obtained allylic silyloxycycles were further oxidized into (Z)-alkenyldiols.

    As a part of our studies on the reactions of 1-ethoxyisochroman (1) with nucleophilic reagents, the reaction of 1 with benzylamines was examined. Heating of 1 with benzylamine or its derivatives having an electron-releasing or-attracting group at the 4-position gave 1-benzylaminoisochroman (2) or the corresponding 1-(4-substituted benzylamino) isochromans (6 and 7). Pyrolysis of 2, 6, and 7 gave 4-benzylisoquinoline (3) and 4-(4-substituted benzyl)-isoquinolines (8 and 9), respectively. Compound 3 was also obtained by heating 2-(2-hydroxyethyl) benzaldehyde with benzylamine, or by heating 2-vinylbenzylidenebenzylamine (4), while the reaction of 1 with N-methylbenzylamine afforded 1-(N-methylbenzylamino) isochroman (5) and did not give the 4-benzylisoquinoline derivative at all. The reaction mechanism giving 4-benzylisoquinolines is proposed to be as shown in Chart 3.

    A novel bilirubin antigen capable of functioning as an antigen which does not undergo decomposition in vivo. This antigen has a structure wherein a protein is bound to the methylene group of the outer vinyl group present in the outermost ring of bilirubin through a spacer or to at least one carboxyl group. A monoclonal antibody capable of recognizing the bilirubin antigen can be obtained from hybridoma cells prepared by cell fusion between antibody-yielding cells obtained by immunizing with the bilirubin antigen and tumor cells capable of division and growth in a culture medium. This monoclonal antibody can recognize bilirubins present in vivo without decomposing their structures, and hence it enables direct qualitative and analyses of bilirubins.
