peptides containing two cysteines are described. These scaffolds contain a functionality with orthogonal reactivity to be used for labeling and ligation. This is illustrated by the chemicalligation of two dissimilar constrained peptides via oxime ligation or strain-promoted azide–alkyne cycloaddition in aqueousmedia.

Nuc 'em: A variety of nucleic acids can be catalytically alkylated with rhodium‐carbenoids generated from diazo compounds in aqueous buffer through an NH insertion process (see scheme; MES=2‐(N‐morpholino)ethanesulfonic acid). The method specifically targets unpaired bases such as those present in single strands, turn regions, and overhangs while leaving double‐stranded sequences untouched.

A simultaneous stereoselective 2-O-deacetylation and 4-amination reaction of peracetylated Neu5Ac 1 has been established with cyclic secondary amines, such as 1-N-Boc-piperazine. Four C-2-symmetric and two asymmetric sialic acid dimers with (4 -> 4)-piperazine derivatives linked were synthesized. They may serve as precursors of unnatural polysialic acids. The invention relates to the fields of peptide mimetics and pharmacy. The invention provides novel cycle forming linkers and bicyclic peptide mimetics prepared therefrom. The linkers comprise an organic moiety P, leaving groups X1 and X2 at benzylic positions, and a reactive group Q capable of participating in a linking reaction. The organic moiety P contains an aromatic (hetero) cycle, an aliphatic heterocycle comprising a positively charged nitrogen atom, and a neutral nitrogen atom.
