Isolation, Structure Determination, and Synthesis of Galaxamide, A Rare Cytotoxic Cyclic Pentapeptide from a Marine Algae Galaxaura filamentosa
摘要:
Galaxamide (1), a rare cyclic pentapeptide, was isolated from the marine algae Galaxaura filamentosa. A preliminary bioassay of Galaxamide showed remarkable in vitro antiproliferative activities against GRC-1 and HepG(2) cell lines. The first total synthesis of the cyclic peptide was achieved for further biological evaluation.
cleavage of caspase-9 and caspase-3, as well as increased intracellular Ca levels and decreased the mitochondrial membrane potential. Collectively, certain linear leucine dipeptides derived from cyclicpentapeptide are able to inhibit tumor cell proliferation through cell cycle arrest and apoptosis induction. The N-methyl group in the side chain and the D/L conformation of the amino-acid residue are critical
