Flexible and practical synthesis of 3-oxyindoles through gold-catalyzed intermolecular oxidation of o-ethynylanilines

A novel gold-catalyzedintermolecular oxidation of o-ethynylanilines has been developed. A range of functionalized 3-oxyindoles are readily accessed by utilizing this strategy. Importantly, this gold-catalyzed oxidative process outcompetes the typical indole formation.
Gold(III)-catalyzed annulation of 2-alkynylanilines in EtOH or EtOH-water mixtures at room temperature gives indoles derivatives in good yields. One-flask protocol for the gold-catalyzed conversion of 2-alkynylanilines to 3-bromo and 3-iodoindoles is also reported.

This study reports geminal amino- and oxy-trifluoromethylation of ortho-ethynyl anilines/phenols by phenCu(III)(CF3)3 (phen denotes 1,10-phenanthroline), which constructs indole and benzofuran skeleton with concurrent incorporation of a CF3 group to the C2 position. Sequential trifluoromethylation of the terminal alkyne followed by 5-endo-trig cyclization of amino/hydroxyl with alkyne is involved.
