Valproicacid (VPA, 1) is a major broad spectrum antiepileptic and central nervous system drug widely used to treat epilepsy, bipolar disorder, and migraine. VPA’s clinical use is limited by two severe and life-threatening side effects, teratogenicity and hepatotoxicity. A number of VPA analogues and their amide, N-methylamide and urea derivatives, were synthesized and evaluated in animal models of
Disclosed is a dihydropyrimido fused ring derivative as a HBV inhibitor, and in particular relates to a compound shown as formula (I) or a pharmaceutically acceptable salt thereof.

The steric limits to the alkylation of aliphatic nitriles and carboxylic acids have been investigated in some detail. For the experimental conditions considered (ionization by i-Pr2NLi in THF followed by alkylation with RI/THF/HMPA) the most hindered nitriles R-CN and carboxylic acids R-CO2H have the same secondary alkyi group RtBuPiCH-, but different tertiary. i.e. Rt-BuPriEtC- or i-Pr3C- for RCN
4-dimethyl-2-oxazolines, 4, after hydrolysis of the intermediate enamine. Alkylation of the latter also permits the synthesis of α,α,α-trisubstituted ketones. α,α-Disubstituted ketones have been prepared by addition of organometallic reagents to 4,4-dimethyl-2-oxazolinium salts, 6. The efficiency of this method has been compared to that of other currently used methods.
