    Design and Synthesis of Lipids for the Fabrication of Functional Lipidic Cubic-Phase Biomaterials
    摘要:
    A series of novel lipids with designed functionalities were synthesized. These lipids are based on conjugation of alpha-amino acids and their esters, cationic, anionic, neutral, and photochromic moieties to the lipophilic 9-cis octadecenyl chains by amide, ester, thioester, or amine bonds. Because of the plasticity of lipidic cubic phases, it is envisaged that when mixed with monooleoyl-rac-glycerol (monoolein, MO) and water at appropriate proportions, they would assemble to form bicontinuous lipidic cubic phases (LCPs) that exhibit the well-known material properties of LCPs such as phase stability, optical transparency, and chemical permeability. Moreover, due to the nature and position of the functionality at the headgroup region, we envision them to perform as functional materials by design.

    A series of novel lipids with designed functionalities were synthesized.     Use of a tryptophanyl-ester or an N-acyl derivative of a tryptophanyl-ester for the prophylaxis and therapy of oxidative pathologic processes in degenerative diseases and/or cancer diseases. Thereby, the tryptophanyl-ester or its N-acyl derivative is preferred to be tryptophanoctyl, N-oleoyl-tryptophanethyl-ester or N-dodecanoyl-tryptophanethyl-ester. Futhermore the tryptophanyl-esters and their N-acyl derivatives are preferred to be used for the treatment and/or prophylaxis of neurodegenerative diseases, cataracts,- neoplastic diseases and/or cardiovascular diseases. In particular, the invention relates to the use of the tryptophanyl-esters and their N-acyl derivatives for the treatment and/or prophylaxis of Alzheimer's disease, Parkinson's disease, apoplectic fit, amyotrophic lateral sclerosis, cancers, arteriosclerosis and/or myocardial infarction. In addition, the invention relates to a pharmaceutical composition comprising a tryptophanyl-ester or an N-acyl derivative of a tryptophanyl-ester.

    Disclosed is an improved method of making polymers. The method utilizes at least one ethylenically unsaturated monomer and at least one polymerizable surface active agent. The polymerizable surface active agent is capable of co-polymerization with traditional monomers and/or polymerization with itself (i.e. homopolymerization) and is preferably substantially completely consumed during the course of the polymerization. Polymers produced by the method of the present invention are well suited for use in coatings, adhesives, sealants, elastomers and the like, as they form stable films, possess excellent adhesion properties and have improved hydrolytic stability characteristics. The present invention also encompasses homopolymeric surface active agents and their use in polymerization reactions.
