The total synthesis of the tremorgenic mycotoxin Verruculogen TR-2 (6) is presented. Selective oxidation of 1 to yield 3 or 4 was achieved using 2. Compound 4 was elaborated to the title compound employing osmium tetroxide.

FtmOx1-catalysis changes from the endoperoxidation to a hydroxylation reaction and leads to dealkylation. In addition, consistent with the dealkylation side-reaction in the COX-like model prediction, the X-ray structure of the FtmOx1•CoII•αKG•7 ternary complex reveals a flip of Y224 to an alternative conformation relative to the FtmOx1•FeII•αKG binary complex. Reaction of indolyldiketopiperazine 5 with 2,3,4,5,6,6-hexachlorocyclohexadien-1-one (25) in dichloromethane/methanol provided the methoxyindolenine 29, which in the presence of TFA rearranged to the dehydrodiketopiperazine 6. This compound was elaborated to verruculogen TR-2 (2) employing osmium tetroxide oxidation.
