The present invention relates to methods for detecting the position of a nucleotide modification, selected from the group consisting of N6-methyladenosine (m6A) and N4-methylcytidine (m4C), in one or more RNA species. Said methods are based on a deamination of the RNA followed by reverse transcription and sequencing of the resulting cDNAs.
