Expedient Synthesis of N-Substituted 2-Aminothiazoles

The reaction of alpha-halo ketones 1 with potassium thiocyanate and amines 3 offers the advantage of an efficient one-pot synthesis of the title compounds 4 from readily available starting materials.
An efficient synthesis of 2,4,5-trisubstituted thiazoles via the reaction of α-nitroepoxides and thioureas under mild conditions has been developed. This reaction proceeded well at room temperature, to afford products in excellent yields for a wide range of substrate, and a possible mechanism has also been proposed.
Abstract Polysubstituted 2-aminothiazoles have been synthesized via a one-pot three-component reaction of α-nitro epoxides, potassium thiocyanate, and primary amines without the need for any additives. This reaction proceeds smoothly in a highly efficient and eco-friendly manner with good to excellent yields. A possible mechanism is also proposed.
