  α-Isocyano-β-phenylpropionamide 1 is synthesized from the corresponding amino acid in excellent yield. The unique reactivity of this bifunctional compound is exploited for the development of novel multicomponent synthesis of 5-aminooxazole 6, pyrrolo[3,4-b]pyridin-5-one 10 and 4,5,6,7-tetrahydrofuro[2,3-c]pyridine 13.

  By simply heating a toluene solution of isocyanoacetamide (3), amine (4), and aldehyde (5), a clean three-component reaction occurred to provide the pyrrolidinone-fused azaindoline (2). In this multicomponent reaction, the isocyanoacetamide (3) reacted four times in a highly ordered manner creating three heterocylic rings with the concurrent formation of five chemical bonds and a minimal loss of molecular weight. Heating is the only external energy required to promote this powerful complexity-generating MCR.

  A novel three-component synthesis of tetrahydrofuro[2,3-c]pyridines (1) from readily accessible starting materials is described. Simply heating a toluene solution of an aminopentynoate (2a), an aldehyde (3), and an alpha-isocyanoacetamide (4) in the presence of ammonium chloride provided the 4,5,6,7-tetrahydrofuro[2,3-c]pyridines (1) in good to excellent yield. The fused ring system is produced in this one-pot process by the concomitant formation of five chemical bonds.
