The reaction of spiroepoxycyclohexadienones 1 with TMSCN in the presence of catalytic amounts of Bu4NCN results in the formation of two diastereomeric cyanohydrins. Alternatively, the reaction of 1 with equimolecular amounts of Bu4NCN gave rise to products arising from two other different reaction paths.

Diels-Alderreaction of cyclopentadiene with spiroepoxycyclohexadienones obtained by Alder oxidation of salicyl alcohols affords mainly endo-endo adducts. Cope rearrangement of normal-electron-demand adducts and endo-endo inverse-electron-demand addition of 2a with ethylvinyl ether are also shown.
Antiangiogenic versus cytotoxic activity in analogues of aeroplysinin-1

A series of analogues of the potentially angiogenic inhibitor aeroplysinin-l 1 were synthesized and their in vitro antiangiogenic and cytotoxic activities evaluated. In the case of epoxy ketone 6 and azlactone 36 the relationship sprouting inhibition assayl cytotoxicity in BAE cells was enhanced by one order and two orders of magnitude, respectively, with respect to the reference. These results imply more specific antiangiogenic properties for the synthesized derivatives. (c) 2007 Elsevier Ltd. All rights reserved.

The invention provides immunostimulatory compositions comprising a small molecule immuno-potentiator (SMIP) compound and methods of administration thereof. Also provided are methods of administering a SMIP compound in an effective amount to enhance the immune response of a subject to an antigen. Further provided are novel compositions and methods of administering SMIP compounds alone or in combination with another agent for the treatment of cancer, infectious diseases and/or allergies/asthma. In a further aspect, the invention relates generally to methods of screening for small molecule immuno-modulatory compositions.
