    The present invention relates to a method for producing cytidine 5'-monophospho-N-acetyl-neuraminic acid (CMP-Neu5Ac, 1) from low-cost substrates N-acetyl-D-glucosamine (GlcNAc), pyruvate, cytidine and polyphosphate in a single reaction mixture with a set of optionally immobilized or optionally co-immobilized enzymes comprising N-acylglucoamine 2-epimerase (AGE), an N-acetylneuraminate lyase (NAL), an N-acylneuraminate cytidylyltransferase (CSS), a uridine kinase (UDK), a uridine monophosphate kinase and a polyphosphate kinase 3 (PPK3). Further, said process may be adapted to produce Neu5Acylated i.e. sialylated biomolecules and biomolecules including a saccharide, a peptide, a protein, a glycopeptide, a glycoprotein, a glycolipid, a glycan, an antibody, and a glycoconjugate, in particular, an antibody drug conjugate, and a carbohydrate conjugate vaccine, or a flavonoid.

    An apheresis column for the treatment of mammals to reduce galectin-3 levels is set forth. The column features, as a stationary or adsorbent phase, pectin molecules, which naturally bind gal-3. Modified citrus pectin, having a molecular weight of about 40 kD or less, preferably about 25 kD or less, is a preferred adsorbent. The columns are disposable, and are used in the fashion for apheresis in general. Other targets, particularly including other galectins, as well as toxins, heavy metals and the like may also be withdrawn from the blood or plasma through this method. Gal-3 level reductions of 10%, 30% and even more may be achieved in a single treatment.

    The present invention provides compositions and methods for inducing an immune response in a subject in need thereof, comprising administering to the subject an immunologically-effective amount of a live Salmonella Typhi vector, wherein the Salmonella Typhi vector has been engineered to express one or more cancer antigens. In some aspects the vector has been engineered to express an outer membrane folding protein Bam A or a fragment or variant thereof; and a lipid A deacylase PagL or a fragment or variant thereof, wherein the Salmonella Typhi vector is capable of delivering the antigen to a mucosal tissue or subcutaneously to dendritic cells via an outer membrane vesicle when administered to a subject.
