Rearrangement of thiazolidine derivatives – a synthesis of a chiral fused oxathiane–γ-lactam bicyclic system

Reaction of L-cysteine with carbonyl compounds leads to thiazolidine derivatives which undergo a stereoselective conversion to two types of chiral bicyclic products bearing two or three stereogenic centers, including the first fused oxathiane–γ-lactam system.
The preparation of thiazolidine-4-carboxylicacidderivativescontaining a 2-nitrooxyethylamine group, potentially active as vasodilators, is reported. Their 1H nmrstudies carried out to establish the configuration of the C2 stereocenter and the full assignment of their 1H and 13C nmr spectra, are also reported.

possible influence on tolerance phenomenon, it was important to check the vasorelaxing effects of our compounds compared to those of commercial organic nitrates of similar structures such as isosorbide mononitrate or isosorbide dinitrate. All the compounds were tested on isolated rat aortas; some of these products exhibited an interesting activity.
