Calcium Induced Calcium Release (CICR) in β-cells has been found to be a novel target for stimulating insulin secretion in a glucose-dependent manner. The present invention relates to a method of identifying compounds that stimulate insulin secretion in a context-dependent manner based on the finding that compounds stimulating insulin secretion in a glucose-dependent manner are able to elicit periodic amplified Ca2+ release in β-cells. The invention also relates to the use of such compounds, for the manufacture of a medicament for use in the treatment of defective insulin secretion related disorders, especially type 2 diabetes.
