Modification of the pyrimidone 5-substituent in a series of 1-((amidolinked)-alkyl)-pyrimidones, lipophilic inhibitors of lipoprotein-associated phospholipase A(2), has given inhibitors of nanomolar potency and improved physicochemical properties.
