N-Heterocyclic Carbene Catalyzed Dynamic Kinetic Resolution of Pyranones

The dynamickineticresolution of 6‐hydroxypyranones with enals or alkynals through an asymmetric redox esterification is catalyzed by a chiral N‐heterocyclic carbene. The resulting esters are obtained in good to high yields and with high levels of enantio‐ and diastereocontrol. The reaction products are further derivatized to obtain functionalized sugar derivatives and natural products.
The present disclosure relates to 2,4-pyrimidinediamines substituted with tricyclic carbamates and the compositions and methods using these compounds in the treatment of conditions in which modulation of the JAK pathway or inhibition of JAK kinases, such as JAK2 or JAK3, is therapeutically useful.

The first carbene-catalyzedregio- and enantioselective indole C7-alkylation reaction between 4-aminoindoles and α-bromoenals is disclosed. The corresponding indole products could be obtained in moderate to good yields with good to excellent enantioselectivities. The evaluation of antibacterial activity against Psa revealed that nine of the C7-functionalized indoles exhibited superior inhibitory activities
