Further transformations afforded 1-(t-butyldimethylsilyloxymethyl)-3(R,S)-hydroxybicyclo[2,1,1]hexanes (11), which were condensed with 6-chloropurine and 4-acetylcytosine via Mitsunobu reactions to give, after further treatment, 3(R,S)-(adenin-9-yl)- and 3(R,S)-(cytosin-1-yl)-1-hydroxymethylbicyclo[2,1,1]hexanes (14 and 17), respectively.
3,3-二(异丙氧基羰基)-1,1-二甲氧基环丁烷(1)经多步转化为3-(1,3-二硫代环己基-2-基)-1,1-二(对甲苯磺酰氧基甲基)进行丁基锂介导的环化反应的环丁烷(7),得到双环[2,1,1]己烷环系统8。进一步的转化得到了1-(叔丁基二甲基甲硅烷基氧基甲基)-3(R,S)-羟基双环[2,1,1]己烷(11),将其通过Mitsunobu反应与6-氯嘌呤和4-乙酰胞嘧啶缩合,在进一步纯化后得到。处理,3(R,S)-(腺嘌呤-9-基)-和3(R,S)-(胞嘧啶-1-基)-1-羟甲基双环[2,1,1]己烷(14和17)。
