A dramatic acceleration of the enantioselective copper-catalyzed conjugatereduction of alpha,beta-unsaturated lactones, lactams, and esters is reported upon addition of alcohol additives. Good to excellent yields and enantioselectivities were realized using a catalyst generated in situ from CuCl(2).H(2)O, t-BuONa, p-tol-BINAP, and PMHS, and this methodology was applied to the synthesis of (-)-Paroxetine

69–92% yield by reductive dehalogenation of α-halo-β-arylbutenolides. The latter were assembled in a single-step from α,β-dihalobutenolides, which are accessible on a large-scale from biomass-derived furfural. Our dehalogenation protocol is illustrated by a new synthesis of the marineantibiotics rubrolide E and F, and 3″-bromorubrolide F.

largely unknown secondary metabolism of the plant pathogenic fungus Hymenoscyphus pseudoalbidus was investigated by use of the CLSA method. A set of volatilelactones was identified by GC/MS. The lactones were synthesized and used in bioassays in which one of the compounds was found to be a strong germinationinhibitor for ash seeds, causing necroses in the plant tissue.
