Synthesis and evaluation of anti-tumor activities of N4 fatty acyl amino acid derivatives of 1-β-arabinofuranosylcytosine

1-beta-D-Arabinofuranosylcytosine (Ara-C, Cytarabine) is one of the drugs used for acute nonlympbocytic leukemia (ANLL). However, the bioavailability of Ara-C is relatively low due to its low lipophilicity. In order to improve the lipophilicity and bioavailability of Ara-C, a series of N-4 derivatives of Ara-C, i.e., (fatty acid)-(amino acid)-Ara-C analogues, were prepared. The 15 derivatives synthesized were characterized by their melting points, optical rotations and partition coefficients. It was found that the Ara-C derivatives synthesized in this study were more lipophilic than Ara-C as determined by their partition coefficients. Their in vitro cytotoxicity and in vivo anti-tumor activity were determined and compared with that of Ara-C. It was found that the derivatives were more active than Ara-C in Hela cells, but not in HL-60 cells. The in vivo results showed that some of the derivatives were more effective than Ara-C in mice bearing S-180 tumor while others showed a decreased activity in comparison with Ara-C. A preparative method is proposed for obtaining higher N-acylamino acids by reaction of free amino acids with fatty acid nitrophenyl esters. It was shown that these acids can transport positive ions through a liquid lipophilic medium. A direct method is proposed for obtaining fatty acid 4-nitrophenyl esters by boiling 4-nitrophenol and the fatty acid in xylene in a Soxhlet apparatus in the presence of an acid catalyst.

Treatment of soy protein with a cation exchange resin greatly enhances the aqueous solubility of the protein without altering the native distribution of protein molecular weights. The resulting protein, in solutions of at least 20% by weight, provides a high quality, soy protein-based meat analog binder which binds at relatively low temperatures, generally above about 55°C but less than 80°C. The binder can be incorporated in shaped, textured protein food products, formed by coating or mixing particulate textured protein material with the binder, shaping the coated particulate material into a unitary shaped product, and heating the shaped product to heat set the binder.
Stable hygroscopic compositions and methods for stabilizing hygroscopic ingredients

The present invention provides methods and compositions for providing controlled protection for hygroscopic ingredients that will be exposed to moist or humid environments for significant periods of time by encapsulating the hygroscopic ingredients with a controlled-protection lipid coating. The methods and compositions are particularly useful for stabilizing a hygroscopic bioactive substance, such as choline chloride or lysine hydrochloride, in an animal feed composition and also providing adequate rumen protection in ruminant feeds.
