Inactivation of Protein Farnesyltransferase by Active-Site-Targeted Dicarbonyl Compounds

Upon farnesylation by protein farnesyltransferase (FTase), key proteins become compartmentalized in cells. For example, cell membrane localization is essential for the mitogenic role of mutant Ras protein, which acts as a switch for cancer cell proliferation. We repel? that alpha -dicarbonyl compounds derived from the isoprenoid skeleton or other hydrophobic groups potently obstruct farnesylation of a Ras model peptide by human recombinant FTase in vitro. A geranyl-derived isoprenoid diketone, 5.9-dimethyl-8-decene-2,3-dione, at 17 muM caused a 62% reduction in FTase activity after 30 minutes. A farnesyl-derived isoprenoid diketone, 5,9,13-trimethyl-8,12-tetradecadiene-2,3-dione, at 93 muM caused a 94% reduction after 30 minutes. Other dicarbonyl:compounds found to be effective against FTase in vitro were (+/-)-6-(camphorquinone-10-sulfonamido)-hexanoic acid, 4,4 ' -biphenyldiglyoxaldehyde, dehydroascorbic acid 6-palmitace, 2-oxododecanal, and phenylglyoxal. Higher concentrations of the alpha -dicarbonyl compound resulted in more rapid and more extensive inactivation. These findings demonstrate that ct-dicarbonyl compounds targeted to FTase interfere with protein farnesylation in vitro and may,lead to derivatives that have utility as chemotherapeutic agents.
There is provided a lubricating composition including a lubricating base containing an asymmetric sulfurized oxymolybdenum dithiocarbamate of the formula (1); a symmetric sulfurized oxymolybdenum dithiocarbamate of the formula (2); and a phenolic or aminic antioxidant.

This lubricating composition meets both the required solubility of additives in base oils and long drain properties concurrently.
In diesel engines, soot is produced by combustion and enters into the engine oil, preventing the engine lubricant from fully exercising its lubricity. The present invention provides a lubricating composition comprising a lubricating basestock, an organic molybdenum compound (A) and succinimide (B) having 0.01 or less of an IR spectrum absorbance peak intensity ratio α/β, wherein α represents an absorbance peak intensity at 1,550±10cm-1 and β represents an absorbance peak intensity at 1,700±10cm-1. Preferably, the organic molybdenum compound is the compound represented by the following formula (1) or formula (2):
(wherein R1 to R8 each independently represent a hydrocarbon, and X1 to X8 each independently represent a sulfur atom or an oxygen atom), or a reaction product of an amine and a molybdenum compound having at least one pentavalent or hexavalent molybdenum atom.

It is to provide fine particles of copper, nickel or palladium hydride having an average particle diameter of at most 50 nm, which are hardly oxidized in the atmosphere and are excellent in storage stability and are thereby very suitable for formation of metallic materials, and their production process. The present invention provides an engine oil composition that contains a base oil containing a low amount of aromatic components and can maintain lubricity because an organic molybdenum compound barely decomposes during long-term use without containing high amounts of an antioxidant or a zinc dithiophosphate compound.
In order to achieve this, the present invention provides an internal combustion engine lubricating oil composition containing a base oil that is a hydrocarbon oil containing less than 1% by mass of aromatic components and having a sulphur content of less than 20 ppm by mass; a specific organic molybdenum compound as a component (A) at 300 to 1500 ppm by mass as molybdenum atoms; and a borate ester compound as a component (B) at 100 to 1000 ppm by mass as boron atoms.
