    We report the first fluorescent diamino-chalcone library and its application in the discovery of a mouse embryonic stem cell (mESC) probe. CDg4, a novel green fluorescent mESC probe was discovered through a high-content image based screening of 160 members of the chalcone library. Interestingly, the molecular binding target of CDg4 was identified as the glycogen of the stem cell colony surface, rather than a conventional protein target from an intracellular source.

    Six nitrones joined by amides to olefins were prepared in situ from the related ketones 1–3 with N-methy]- and benzylhydroxylamines. The nitrones added intramolecularly to the olefins, and the cycloadditions gave the 6-lactams 4–9 stereoselectively and regiospecifically.     1-(2-Cyclohexenyl-1-carbonyl)-piperidine-2-carboxaldehyde oxime (23) was prepared in 5 steps from pyridine-2-carboxaldehyde. Oxidation with NaOCl afforded the corresponding nitrile oxide which cyclized to give a 1:1 ratio of diastereomers 2a alpha,3,4,5,5a alpha,9,10,11 alpha,beta,11c alpha-decahydro-6H,8H-[1,2] benzisoxazolo [3,4-ab] quinolizin-6-one (24A) and 2a alpha,3,4,5,5a alpha,9,10,11a alpha,11c alpha-decahydro-6H,8H-[1,2] benzisoxazolo [3,4-ab] quinolizin-6-one (24B) in a best yield of 30%.
